Cryopreservation of Pacific oyster ( Crassostrea gigas ) larvae: Revisiting the practical limitations and scaling up the procedure for application to hatchery ArchiMer
Labbe, Catherine; Haffray, Pierrick; Mingant, Christian; Quittet, Benjamin; Diss, Blandine; Tervit, H. Robin; Adams, Serean L.; Rimond, Flore; Suquet, Marc.
Pacific oyster Crassostrea gigas is one major species for aquaculture, and the development of breeding programs and the need for preservation of wild stock genetic resources prompted the need for larvae cryopreservation. The objective of the present study was to choose the most reliable protocol from several existing publications, to test its biological and practical limitations, and to adapt it to hatchery conditions. The selected protocol was characterized by a very slow freezing rate without seeding, and by the use of ethylene glycol and sucrose as cryoprotectant. The best survivals after thawing and rearing up to 48 h post fertilization (hpf) were obtained with larvae that were frozen at late trochophore (20 hpf) and early-D (24 hpf) stages. Increasing...

Effects of cryoprotectant and plunging temperature in liquid nitrogen on the in vitro and in vivo development of murine morulae Braz. J. Vet. Res. Anim. Sci.
PANTANO,Thaís; MELLO,Marco Roberto Bourg de; GARCIA,José Fernando; HO,Linda Lee; VISINTIN,José Antonio.
The effects of plunging temperature in liquid nitrogen and cryoprotectant dilution methods were evaluated for compacted mouse morulae frozen in 1.5 M ethylene-glycol (E), 1.5M propylene-glycol (P) or 1.4M glycerol (G). Morulae were equilibrated for 10 minutes in cryoprotectant solution and loaded into 0.25 ml straws with cryoprotectant solution in 3 columns (groups E1, P1, G1) or cryoprotectant in the center and PBS in the lateral columns (E2, P2). Straws were cooled at 0.5ºC/min to -25 or -30ºC and plunged into liquid nitrogen. Straws were thawed in water at 22ºC for 20 seconds. Cryoprotectant was diluted in 3 steps for group G1 and in one step for groups E1 and P1 (direct transfer to PBS + 10% FCS) and E2 and P2 (shaken to mix the 3 columns before...

In vitro evaluation of canine spermatozoa cryopreserved in different extenders Arq. Bras. Med. Vet. Zootec.
Oliveira,E.C.S.; Juliani,G.C.; Marques Jr.,A.P.; Henry,M..
The efficacy of three extenders, tris-egg yolk-5% ethylene glycol (T1), lactose-egg yolk-5% ethylene glycol (T2) and lactose-egg yolk-5% dimethyl formamide (T3) on preserving the viability of post-thawing canine spermatozoa was evaluated. Three ejaculates per dog were obtained of five animals. The semen was packaged in 0.5ml straws and cooled to 4°C for 120min. The straws were frozen 4cm above the nitrogen level for 15min and thawed in water-bath at 37°C for 60sec and at 75°C for 7sec. Progressive motility and vigour were evaluated immediately after thawing (time 0) and at 30, 60, 90 and 120min. Structural and functional integrity of plasma membrane of the spermatozoa were evaluated, respectively, by fluorescent staining probes and hypoosmotic swelling...

In vitro viability of canine spermatozoa frozen in tris-fructose-citric acid extender with ethylene glycol Braz. J. Vet. Res. Anim. Sci.
VANNUCCHI,Camila Infantosi; SANTOS,Silvia Edelweiss Crusco dos; VISINTIN,José Antonio.
The use of frozen semen has become increasingly popular in canine artificial insemination, stimulating research to improve freezing methods and extenders. In this study, was compared the final concentration of 5% ethylene glycol used in 3 equilibration protocols. In Method I, one part of semen was added to two parts of tris-fructose-citric acid with 7.5% ethylene glycol (Extender 2) and frozen without cooling. In Method II, one part of semen was added to two parts of tris-fructose-citric acid with 7.5% ethylene glycol (Extender 2) and cooled at 5ºC for 1 hour before freezing. In Method III, one part of semen was added to one part of tris-fructose-citric without ethylene glycol (Extender 1) and cooled at 5ºC for 1 hour. Just after this period, one part of...

Production of lignocellulolytic enzymes by Aspergillus niger biofilms at variable water activities Electron. J. Biotechnol.
Villena,Gretty K; Gutiérrez-Correa,Marcel.
Lignocellulolytic enzyme production by Aspergillus niger was compared both in submerged fermentation (SF) and biofilm fermentation (BF) at varying water activities. Maximal filter paper activity, endoglucanase and xylanase activities were much higher in BF (2.96, 4.7 and 4.61 IU ml-1, respectively) than in SF cultures (1.71, 1.31 and 2.3 IU ml-1, respectively) but biomass yields were lower in BF than in SF (0.338 g g-1 and 0.431 g g-1, respectively). In the presence of 20% ethylene glycol (a w = 0.942) the enzyme activities decreased in both systems but BF still had higher levels (1.0, 1.0 and 2.6 IU ml-1, respectively) than SF cultures (0.6, 0.7 and 1.5 IU ml-1, respectively). An increase in xylanase specific activity of more than 2 fold (from 4.2 to 10.2...

Teratogenic Effect of Ethylene Glycol-Methyl Cellosolve Mixture in Rats.: I. Reproductive Damage International Journal of Morphology
Arteaga-Martínez,Manuel; García-Peláez,Isabel; Aguirre-Luna,Oswaldo M; Saavedra-Ontiveros,Dolores.
We investigated the reproductive damage and teratogenic effect of an ethylene glycol-methyl cellosolve mixture on gestating Wistar rats, which received a daily intraperitoneal dose of different concentration of the mixture on day 8 of gestation until day 20. In rats treated with the mixture the number of live fetuses decreased and reabsorptions increased with increasing concentrations of the mixture, as well as the number of abnormal fetuses. We conclude that the ethylene glycol-methyl cellosolve mixture possesses a higher teratogenic potential than each of its constituents separately, producing reproductive damage, external fetal abnormalities, growth delay, and increased fetal death.

Teratogenic Effect of Ethylene Glycol-Methyl Cellosolve Mixture in Rats: II. Craniofacial and Limb Abnormalities International Journal of Morphology
García-Peláez,Isabel; Aguirre-Luna,Oswaldo M; Saavedra-Ontiveros,Dolores; Arteaga-Martínez,Manuel.
We investigated the teratogenic effect of an ethylene glycol-methyl cellosolve mixture on gestating Wistar rats, that received a daily intraperitoneal dose of different concentration of the mixture on day 8 of gestation until day 20. Multivariate analysis and Post-Hoc Bonferroni tests were used and relative risk and attributable fraction were calculated. In rats treated with the mixture the number of live fetuses decreased and reabsorptions increased with increasing concentrations of the mixture, as well as the number of abnormal fetuses. Abnormalities consisted mainly in atypical craniofacial morphology, protruding tongue, edema, signs of growth delay and shorter limbs, their frequency and severity increased at higher concentrations of the mixture. We...
